No. ExoQuick®-TC UltraPure requires no ultracentrifuge, no pre-concentration equipment, and no ultrafiltration system. The entire workflow is completed using standard laboratory benchtop equipment in under 20 minutes of hands-on time per reaction.

Yes. TEM confirms that vesicles retain their native bilayer membrane morphology after isolation. Functional validation using copGFP-loaded EVs from the XPACK530 stable HEK293 cell line demonstrates that engineered EV cargo is fully preserved through the isolation workflow, confirming suitability for cargo delivery and functional studies.

EVs isolated with ExoQuick®-TC UltraPure are validated for:

  • Nanoparticle Tracking Analysis (NTA)
  • Transmission Electron Microscopy (TEM)
  • Western blot (EV marker detection)
  • ELISA
  • RNA extraction, small RNA/miRNA sequencing, and next-generation sequencing (NGS) — compatible with SBI's NGS service
  • Mass spectrometry proteomics (in-gel digestion)
  • EV uptake and cargo delivery assays
  • In vivo / ex vivo administration studies

ExoQuick®-TC UltraPure outperforms both SEC (e.g., qEV) and tangential flow filtration (TFF) in yield and purity for cell culture media applications. vs. SEC (qEV): Delivers more than 50× higher EV yield, with superior removal of albumin, IgG, and lipoproteins. SEC requires pre-concentration of conditioned media before loading — ExoQuick®-TC UltraPure processes your sample directly, no pre-concentration required. vs. TFF: At equivalent large-volume inputs (100 mL), EQ-TC UltraPure produces significantly cleaner EV preparations — confirmed by Ponceau S staining and Western blot — with higher EV marker expression and lower protein contamination. Unlike TFF, no specialised tangential flow filtration system is... Read more

Yes. ExoQuick®-TC UltraPure is validated for use with both standard tissue culture flask media and large-scale bioreactor harvests. Unlike SEC and TFF, it requires no pre-concentration and no ultrafiltration — making it well-suited for therapeutic EV development and cell therapy production workflows where maintaining yield and purity at scale is critical.

Yes, with one important distinction. The kit is fully compatible with in-gel digestion workflows (trypsin, pepsin) because PEG — the precipitation reagent — does not migrate into the gel and does not interfere with MS analysis. For in-solution digestion workflows, PEG can cause ion suppression and background signals. We recommend consulting with your proteomics facility or contacting our technical team before proceeding with in-solution digestion. For EV proteomics using precipitation-based methods, we also recommend reviewing our application note on EV precipitation vs SEC for proteomics... Read more

The dual-action workflow removes three major contaminant classes: albumin (BSA), IgG, and lipoproteins. Near-complete lipoprotein depletion is confirmed by ApoB ELISA. Albumin and IgG removal is validated by Western blot and Ponceau S staining.

The Small Kit (EQ-TC UltraPure_S) processes 10 mL of conditioned media per reaction. The Large Kit (EQ-TC UltraPure_L) processes 100 mL per reaction. Both kits provide 10 reactions. For scale-up beyond 100 mL — including 1 L bioreactor harvests — contact our technical support team for guidance and bulk options.

Transfected exosomes can be added to the cells in a culture plate grown in exosome-depleted FBS media. The quantity can be adjusted depending on your experimental requirements.
on Jun 28, 2023
Exo-Fect has been successfully used to transfect exosomes with small RNAs, longer mRNAs, and plasmid DNA. Cells treated with these transfected exosomes have demonstrated successful uptake and expression of these nucleic acids.
on Jun 28, 2023
The transfected exosomes can be added to any target cells, including those used in cell culture, depending on your experimental requirements.
on Jun 28, 2023
Although only in vitro data has been established and no internal data is available for in vivo experiments, it's possible that the transfected exosomes may be used for in vivo studies.
on Jun 28, 2023
The kit comes in two sizes, each containing Exo-Fect Reagent, Positive Control siRNA (TX-Red), and ExoQuickTC. The specific quantities differ depending on whether you get the 10 reaction or 20 reaction kit.
on Jun 28, 2023
The Exo-Fect transfection protocol takes less than an hour to complete.
