Q:
How are the shRNA libraries quality controlled??
A:
SBI's QC of our shRNA libraries is to transform bacterial cells with the pooled plasmid library and then select and sequence more than 20 randomly selected clones. We typically obtain less than 10% concatemeric inserts and greater than 90% shRNA inserts.
Q:
How many shRNAs are designed per messenger RNA target?
A:
The libraries are designed in such a way so that there are 3 to 5 shRNA for each target gene. So there is a lot of redundancy built into the library, thus a 50% unique shRNA representation will yield reliable results.
