Applications of SBI's Bioluminescent Imaging Vectors

Molecular imaging with firefly luciferase, fluorescent proteins (GFP/RFP), and thymidine kinase provides a non-invasive and accurate method to track cell fate after cell transplantation in small animals. SBI has created a molecular tool set based on well-establish protocols published on Nature Protocols (Sun N, et al. Nat Protoc. 2009;4(8):1192-201.) and has expanded on this product line to include several different promoter and marker combinations as well as vector formats.

Lentivector-based imaging vectors
SBI's lentivector-based imaging systems offer safe, stable and effective gene delivery in virtually all mammalian cells in tissue culture and in whole animal models. Ex vivo BLI of undifferentiated double fusion (DF) human ES cells (H9 line) are shown in the left panel. The implanted ES cells were tracked for in vivo kinetics of transplanted DF human ES (hES) cells and DF hES cell-derived cardiomyocytes with BLI. Representative images from a single animal receiving intramyocardial injection of 1 x10^6 undifferentiated DF hES cells (upper right panel) or DF hES-derived cardiomyocytes (lower right panel) are shown.

Minicircle-based imaging vectors
SBI also offers non-viral and non-integration solutions for molecular imaging using minicircle technologies developed at Stanford. Minicircle-based imaging vector systems offer safe and efficient gene delivery in vitro and in vivo. The Minicircle DNAs are devoid of any bacterial plasmid DNA backbone and their smaller molecular size enables more efficient transfections and offers sustained expression over a period of weeks or months. Minicircle DNAs are also less toxic to cells with lowered stress and inflammation responses. The data below show the transfection efficiency of minicircle in vitro and in vivo. HL-1 cells were transfected with MC-Fluc-GFP (MC-LG) in increasing amounts of Minicircle DNA as indicated in a 6-well plate. The left panel shows bioluminescence imaging analysis of 25 ug Minicircle DNA indicates minicircle plasmid-mediated gene expression was stable for at least 8 weeks in the heart. See Hu S, et al.
