Validation Data for pGreenFire1-STAT1 Transcription Reporter

● Monitoring STAT1 transactivation using GFP epifluorescence

(Interferon-alpha = control experiment; Interferon-gamma = positive control)

 ● Quantitative analysis of STAT1 transactivation using Firefly Luciferase

(Luciferase assays measured using the exact cells from the GFP epifluorescent studies shown above)

293TN cells were transduced with packaged pGF1-STAT1 reporter virus. Two days later, the cells were split into 96 well plates and then treated with the indicated amount of Interferon-alpha or -gamma.  STAT1 activity levels were assessed 18 hours post-induction using epifluorescence microscopy and a standard firefly luciferase assay (Promega).
 

Kit Components

Each kit contains 10 μg of endotoxin-free plasmid DNA in TE buffer or >2 x 10⁶ IFUs pre-packaged lentivirus.

Each kit comes with a product certificate of analysis.
