pGreenFire1-ISRE (Interferon-stimulated Response Element) is an HIV based lentivector that co-expresses destabilized copGFP and Firefly Luciferase in response to transcriptional activation by STAT1/STAT2.

Validation Data for pGreenFire1-ISRE Transcription Reporter

 ● Monitoring ISRE transactivation using GFP epifluorescence

 ● Quantitative analysis of ISRE transactivation using Firefly Luciferase

(Luciferase assays measured using the exact cells from the GFP epifluorescent studies shown above)

293TN cells were transduced with packaged pGF1-ISRE reporter virus. Two days later, the cells were split into 96 well plates and then treated with the indicated amount of Interferon-alpha or -gamma.  STAT1/STAT2 activity levels were assessed 18 hours post-induction using epifluorescence microscopy and a standard firefly luciferase assay (Promega).
 

Kit Components

Each kit contains 10 μg of endotoxin-free plasmid DNA in TE buffer or >2 x 10⁶ IFUs pre-packaged lentivirus.

Each kit comes with a product certificate of analysis.
