Introduce Single or Multiple Mutations Simultaneously
The MetaMorph kit allows for multiple mutations to be made all at once through primer design guidelines (see User Manual). The positive control reaction that comes with the kit creates mutagenic sites in the control plasmid that can be checked by digestion and gel analysis.
Control and Mutated samples cut with NdeI and BamHI. Control vector has only 1 NdeI and 1 BamHI restriction site. The single insertion mutation adds a new NdeI site. The double mutation (insertion and point mutation) adds NdeI and BamHI sites.
