Lentiviral vectors are complexed with magnetic nanoparticles, which are then placed within a strong magnetic field to rapidly pull the virus particles towards the target cells. In this manner, the applied viral dose is concentrated onto cells within minutes so that most of the cells come into contact with the lentiviral vector inoculum. As a result, the number of transduced cells is increased by up to 3-fold as compared to traditional transductions performed using polybrene (Fig. 1 a&b).
LentiMag™ reagent requires a strong magnetic field to enhance transduction of target cells with lentiviral vectors. The magnetic plate supplied with the kit has been specifically designed to exert this magnetic field. The geometry of the plate is compatible with all standard cell culture dishes, including T-25 and T-75 flasks, 60 and 100 mm diam. dishes, and 6-, 12-, 24-, 48-, & 96-well plates.
